FUNCTION: SwissProt: P06493 # Plays a key role in the control of the eukaryotic cell cycle. It is required in higher cells for entry into S-phase and mitosis. p34 is a component of the kinase complex that phosphorylates the repetitive C-terminus of RNA polymerase II.

SUBUNIT: Forms a stable but non-covalent complex with a regulatory subunit and with a cyclin. Interacts with DLG7. Isoform 2 is unable to complex with cyclin B1 and also fails to bind to the CDK inhibitor p21. Interacts with catalytically active CCNB1 and RALBP1 during mitosis to form an endocytotic complex during interphase.

Evaluated by Kinase assay: 10 µl of CDK1/CyclinB1 was mixed with 5 µl Histone H1 substrate (1 mg/ml) and 5 µl water. Reaction was initiated by adding 5 µl of [33P]-ATP cocktail and incubated at 30°C for 15 min. Reaction was read by adding 20 µl to P81 paper and read in the presence of scintillation fluid in a scintillation counter.
