pCDF-1b DNA - Novagen

pCDF-1b质粒具有共表达能力以及表达具有N端His•Tag的融合蛋白的能力，该融合蛋白在纯化和切割后会产生天然蛋白。该质粒具有源自CloDF13（1）和链霉素/大观霉素抗性的来源，可以选择与许多其他基于Novagen T7的表达载体共表达。

该载体包含T7启动子、lac操纵子、核糖体结合位点(rbs)、氨基末端His•Tag编码序列和多个克隆位点(MCS)区域，旨在允许以最小的载体编码融合产生靶蛋白。Pml I克隆位点允许直接融合到His•Tag序列上，以插入钝的插入片段并在适当的阅读框中。对于需要可移动的氨基末端His•Tag序列的应用，MCS包括PshA I克隆位点(GACNNNNGTC)。PshA I位点与肠激酶(EK)蛋白酶（AspAspAspAspLys）的切割位点重叠。将适当设计的插入片段克隆到该位点可重新创建完整的EK位点，并允许通过EK消化去除所有氨基末端载体编码的序列。MCS的其余部分编码在许多其他Novagen表达载体中发现的限制性酶切位点，以促进插入片段转移。可选的C末端S•Tag编码序列，可用于纯化、检测和定量（2)。
