Origami 2菌株在谷胱甘肽还原酶(gor)和硫氧还蛋白还原酶（trxB）中具有突变，从而促进了适当的二硫键形成。

Origami 2宿主菌株是K-12衍生物，在硫氧还蛋白还原酶（trxB）和谷胱甘肽还原酶(gor)基因中均具有突变，从而大大增强了大肠杆菌中二硫键的形成。Origami2菌株对卡那霉素敏感；与最初的Origami菌株一样，gor突变仍然由四环素选择。为了减少分子之间形成二硫键的可能性，仅建议将含有trxB和gor突变的菌株用于表达需要二硫键形成才能正确折叠的蛋白质。

DE3表明宿主是λDE3的溶原菌，因此，在lacUV5启动子的控制下携带T7 RNA聚合酶基因的染色体拷贝。这样的菌株适用于通过用IPTG诱导从克隆在pET载体中的靶基因生产蛋白质。
