General description

Packaging protein 1 (UniProt: P03271; also known as Packaging protein IVa2) is encoded by the IVa2 gene (Gene ID: 5739974) in human. Adenovirus 5. Adenovirus 5 belongs to the Adenoviridae family of non-enveloped viruses that are characterized by linear, double-stranded DNA genomes. Their capsid is an icosahedron that contains at least 12 different viral proteins. Adenovirus 5 is commonly used for gene transfer experiments due to its ability to infect a wide group of different cell types. It can harbor large genes in its genome incorporated via homologous recombination techniques. Packaging protein IVa2 is a multifunctional, homodimeric, protein that is expressed in the intermediate phase of the viral replicative cycle. It is detected in both empty and mature virus particles. It is a component of the packaging machinery that encapsidates the viral DNA into preformed capsids and transcriptional activator of the viral major late promoter. Its DNA-binding region is localized in amino acids 440-449. It binds, along with packaging proteins 2 and 3, to the specific packaging sequence on the left end of viral genomic DNA and displays ATPase activity thereby providing the power stroke of the packaging machinery. Its activity is reported to be stimulated by protein 33K that acts as a terminase. It is shown to specifically bind to the 5′-CG-3′ nucleotides of the repeats making up the packaging sequence. Quality Control Testing

Evaluated by Western Blotting in lysate from HeLa cells infected Human adenovirus type 5.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Adenovirus 5 Packaging protein IVa2 in lysate from HeLa cells infected with human adenovirus type 5, but not in lysate from uninfected cells.
