General description
3-PGDH (PHGDH) protein in an important enzyme in L-serine biosynthesis. 3-PGDH performs the first rate limiting step in the phosphorylated pathway of serine biosynthesis. 3-PGDH is also a biomarker in certain cancers that over express 3-PGDH. Moreover, 3-PGDH is the essential enzyme needed for cells to traverse the S phase and divide, and it plays a key role in T cell activation and proliferation for instance. Interestingly, even though 3-PGDH knockdown and reduced serine synthesis slows growth and proliferation of many cancer cells in vitro 3-PGDH knockdown does not affect tumor maintenance and growth in established breast cancer xenograft models, suggesting that 3-PGDH-dependent cancer cell growth may be context-dependent.
