General description

Paired box protein Pax-6 (UniProt: P26367; also known as Aniridia type II protein, Oculorhombin, Pax-6) is encoded by the PAX6 (also known as AN2) gene (Gene ID: 5080) in human. Pax6 is a transcription factor that has important functions in the development of the eye, nose, central nervous system and pancreas. It is shown to be essential for the differentiation of pancreatic islet alpha cells. It has two highly conserved DNA binding domains, a paired domain and a homeodomain that is followed by a proline, serine, and threonine-rich trans-activation domain. Its C-terminal peptide region is shown to modulate DNA binding by the homeodomain. It is expressed in the developing eye and brain and in retina it peaks at fetal days 51-60. Pax6 competes with PAX4 in binding to a common element in the glucagon, insulin, and somatostatin promoters. Pax6 is reported to regulate specification of the ventral neuron subtypes by establishing the correct progenitor domains. Mutations in PAX6 gene are known to cause Aniridia, a disorder characterized by complete absence of the iris or extreme iris hypoplasia.
