General description
SLAM family member 6 (UniProt: Q96DU3; also known as Activating NK receptor, NK-T-B-antigen (NTB-A), CD352) is encoded by the SLAMF6 (also known as KALI) gene (Gene ID: 114836) in human. SLAMF6 belongs to the 7-member signaling lymphocytic activation molecule family (SLAMF) of receptors broadly expressed in hematopoietic cells. SLAM receptors triggered by homo or heterotypic cell-cell interactions are modulating the activation and differentiation of a wide variety of immune cells and thus are involved in the regulation and interconnection of both innate and adaptive immune response. SLAMF members carry one or more copies of immunoreceptor tyrosine-based switch motif or ITSM (T-X-Y-X-X-[VI]) in their cytoplasmic tails. Upon phosphorylation on the tyrosine residue, ITSM mediates the recruitment of SH2 domain-containing signaling molecules such as SLAM-associated protein (SAP), including SAP in T-, NK, and NK T-cells, EAT-2A and EAT-2B (murine) in NK cells and APCs. SLAMF6/NTB-A is predominantly localized to the cell membrane and is a single-pass type I membrane protein and is synthesized with a signal peptide sequence (aa 1-21), the removal of which yields the mature SLAMF6 with an extracellular domain (aa 22-226), a transmembrane domain (aa 227-247) and, a cytoplasmic domain (aa 248-331). Its expression is primarily observed in cells of the immune system. SLAMF6/NTB-A is a receptor on the surfaces of human NK, T, and B cells, mediating a signal whose malfunction may be involved in X-linked lymphoproliferative disease. Studies show that NTB-A is its own ligand and homophilic interaction of NTB-A enhances NK cell cytotoxicity and influences NK cell proliferation and IFN- secretion. It is also suggested that NTB-A is an interlymphocyte signaling molecule, which serves to orchestrate the activities of immune cells. Dysregulation of SLAMF6 signaling is associated with autoimmune conditions such as systemic lupus erythematosus (SLE), where it can contribute to abnormal immune cell activation.
