Proline-rich transmembrane protein 1 (UniProt: Q6MG82; also known as Dispanin subfamily D member 1, DSPD1, Synapse differentiation-induced protein 4) is encoded by the Prrt1 (also known as Ng5) gene (gene ID: 406167) in rat. DSPD1 is a highly conserved, multi-pass membrane protein of the CD225/Dispanin family that is expressed in the brain and is detected in excitatory synapses. It contains two transmembrane helices, with both the N and C-termini oriented towards the outside of the cell. It plays an important role during synapse development in regulating AMPA receptor (AMPAR) and PSD-95 content at excitatory synapses. It is a component of the outer core of AMPAR complex. AMPAR complex consists of an inner core that is made of four pore-forming GluA/GRIA proteins and four major auxiliary subunits arranged in two-fold symmetry. The inner core of AMPAR complex is complemented by outer core constituents binding directly to the GluA/GRIA proteins at sites distinct from the interaction sites of the inner core constituents. The proteins of the inner and outer core serve as a platform for other, more peripherally associated AMPAR constituents.
