General description

Sulfhydryl oxidase 1 (UniProt: O00391; also known as EC number EC:1.8.3.2, hQSOX, Quiescin Q6) is encoded by the QSOX1 (also known as QSCN6, UNQ2520/PRO6013) gene (Gene ID: 5768) in human. QSOX1 is a single-pass membrane glycoprotein that is expressed is observed in heart, placenta, lung, liver, skeletal muscle, pancreas. It is synthesized with a signal peptide (aa 1-29), which is subsequently cleaved off to produce the mature protein. Its thioredoxin domain is localized in amino acids 36-156. Two main redox-active domains of QSOX1 have been described. The amino terminus interacts with the thiol-containing substrates and the carboxy terminus catalyzes reduction of molecular oxygen. It oxidizes thiols during protein folding, reducing molecular oxygen to hydrogen peroxide. It introduces disulfide bonds into proteins with a high efficiency and prefers protein substrates rather than small mono- and di-thiol containing molecules. It is shown to be essential for proper assembly of extracellular matrix (ECM). Its higher expression is observed in several malignancies, especially breast and pancreatic carcinoma and it is a key enzyme involved in a tumor′s ability to modify the ECM that enables tumors to communicate with and/or modify its environment at the tumor-stroma interface, resulting in invasion and subsequent growth of the tumor. Two isoforms of QSOX1 have been described that are produced by alternative splicing. (Ref.: Lake, DF., and Faigel, DO. (2014). Antioxid. Redox Signal. 21(3); 485-496; Grossman, I., et al. (2012). J. A recombinant fragment corresponding to 517 amino acids of human QSOX1 protein.

Application

Quality Control Testing

Evaluated by Immunocytochemistry in MDA-MB-231 cells.

Immunocytochemistry Analysis: A 1:1,000 dilution of this antibody detected QSOX1 in MDA-MB-231 cells.

Preparation Note

0.5 mg/mL. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
