Application
Anti-PELI3 (AB2) antibody produced in rabbit is suitable for western blotting at a concentration of 1μg/mL.
Biochem/physiol Actions
The gene PELI3 encodes a scaffold protein involved in TLR (Toll-like receptor)/IL-1R (IL-1 receptor) signalling pathways that shape innante immunity via interaction with the complex containing IRAK kinases and TRAF6. This leads to to nuclear factor-κB and mitogen activated protein kinase-dependent gene expression. The pellino protein also induces IRAK-1 polyubiquitination in a RING-dependent manner, which in turn regulates TLR/IL-1R signalling. Pellino3 targets the interferon regulatory factor 7 pathway and facilitates autoregulation of toll-like receptors (TLR3)- and viral-induced expression of type I interferons. It also plays a role in mediating ′Lys-63′-linked polyubiquitination of IRAK1 and can stimulate AP1/JUN and ELK1.

Members of the Toll-like receptor (TLR) and IL-1 receptor (IL-1R) family initiate signalling pathways that shape innate immunity. Pellino proteins have recently been implicated as evolutionary conserved scaffold proteins in TLR/IL-1R signalling leading to nuclear factor-kappaB and mitogen activated protein

Toll-like receptors (TLRs) sense pathogen-associated molecules and respond by inducing cytokines and type I interferon. Here we show that genetic ablation of the E3 ubiquitin ligase Pellino3 augmented the expression of type I interferon but not of proinflammatory cytokines in

Members of the Pellino family are interleukin-1 receptor-associated kinase (IRAK)-interacting proteins that possess RING-like domains. The presence of these domains led to the suggestion that Pellino proteins are ubiquitin-protein isopeptide ligases (E3). However, no conclusive data currently exist to prove
