Biochem/physiol Actions
Mocha is a naturally occuring mouse mutant with a null mutation in the delta subunit of AP-3 complex. In Mocha cells, the β3 amd μ3 subunits coassmeble into a heterodimer whereas the sigma 3 subunit remains monomeric. The mouse has a similar phenotype to the human genetic disorder, Hermansky Pudlak Syndrome (HPS). In both mice and humans, AP-3 deficiency results in hypopigmentation because of abnormalities in melanosomes. The Mocha cell line can be used to investigate the assembly of AP-3 complexes and AP-3 function.

Preparation Note
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:10 to 1:20 i.e. seeding at 5x1,000 to 1x10,000 cells/cm2 using 0.25% trypsin/EDTA; 5% CO2; 37°C.
