Rat Bronchial Epithelial Cells (RaBE)
Spontaneously immortalised rat bronchial epithelial cell line derived from a 4 month old male Wistar rat. The cell line is basal cell-like expressing cytokeratins 14 and 15. Vimentin negative. A slow growing derivative of RaBE, cultured in the absence of growth factors is also available - RaBE (Slow Growth), Sigma Catalogue number. 04100602.
Preparation Note
Ham′s F12 + 2mM Glutamine + 10ug/ml transferrin + 5ug/ml insulin + 10ng/ml EGF + 10ng/ml aFGF + 100ng/ml cholera toxin + 5% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) between 1:100 and 1:800 i.e. seeding 5000 cells/25cm2 with 5ml or 10,000 cells/75cm2 with 20ml using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C. Subculture once a week, double volume
