Established from peripheral blood of a 48 year-old male with widespread subcutaneous nodules. Biopsy showed high grade Non-Hodgkin lymphoma with numerous mitoses.

Preparation Note
Doubling time approximately 60 hours. Cells grow slowly in suspension in large clumps. Maintain cultures between 3-9 x 105 cells/ml; 5% CO2; 37 °C. Thaw ampoule and perform a cell count, spin out the DMSO and resuspend in an appropriate volume of media to give a cell concentration of 4-5 x 105 cells/ml. Viability may be poor on resuscitation and may decrease further within the first 48 hours, the culture may appear non-viable at this time. Clusters of cells will eventually appear but this could take 1-2 weeks, during this time large media changes are not recommended. It may take 2-3 weeks for vigorous growth to be established. At subculture perform a cell count and maintain cultures between 3-9 x 105 cells/ml, 5% CO2, 37 °C. For cryopreservation of KARPAS 384, freeze at approximately 2 x 107 cells/ml in 90% FBS + 10% DMSO.
