Sub-line of BUdR resistant strain of the L-M mouse line (Sigma Catalogue number. 87032401), which in turn was derived from L929 (Sigma Catalogue number. 85011425). TK- cells cannot grow in HAT medium. Stable TK- mutants are only maintained if the cells are kept for several passages in growth medium containing BUdR.
Preparation Note
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split semi-confluent cultures seeding at 5x1,000 to 2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
