Biochem/physiol Actions
HeLa DH cells are the wildtype that has been used for connexin-transfection assays and subsequent analysis of permeability and gap junction formation. The wildtype exhibits stable membrane potentials, very low gap junctional conductance and high stability under double whole-cell patch-clamp measurements. Gap junction plaques were not detected but using antibody directed against connexin 45 identified gap junction channels as dispersed particles in non typical gap junction areas.

Preparation Note
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Split sub-confluent cultures (70-80%) 1:3 to 1:8 i.e. seeding at 1-3x10,000 cell/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
