General description
DDRGK domain-containing protein 1 (DDRGK1) is an endoplasmic reticulum (ER)-associated protein that is stimulated by ER stress. This protein has a signal peptide for ER anchoring, a transmembrane helix and a proteasome component (PCI) domain. DDRGK1 is located on human chromosome 20.

Biochem/physiol Actions
DDRGK domain-containing protein 1 (DDRGK1) could create a large protein complex with UFL1/NLBP (Ufm1-specific ligase 1), the putative tumor suppressor C53/LZAP and Ufm1 (ubiquitin-fold modifier 1), that participates in ufmylation. It maintains endoplasmic reticulum (ER) homeostasis. The PCI domain of DDRGK1 serves as a protein-protein interaction mediator, that participates in various multi-protein complexes that modulate the protein lifespan.
