General description
Opioid related nociceptin receptor 1 (OPRL1) is mapped to human chromosome 20q13.33. The gene codes for the nociceptin/orphanin FQ receptor (NOP-R), which is a G protein-coupled receptor expressed at various levels all over the brain and spinal cord. OPRL1 belongs to the opioid receptor family.
The protein encoded by this gene is a G protein-coupled receptor whose expression can be induced by phytohemagglutinin. The encoded integral membrane protein is a receptor for the 17 aa neuropeptide nociceptin/orphanin FQ. This gene may be involved in the regulation of numerous brain activities, particularly instinctive and emotional behaviors. A promoter for this gene also functions as a promoter for another gene, regulator of G-protein signalling 19 (RGS19), located on the opposite strand. Two transcript variants encoding the same protein have been found for this gene. (provided by RefSeq)

Biochem/physiol Actions
Opioid related nociceptin receptor 1 (OPRL1) plays a vital role in alcohol and other drug reward pathways. The encoded protein is involved in regulating tolerance and behavioral responses to morphine. Phosphorylation of NOPR at serine 363 by G protein-coupled receptor kinase 3 (GRK3) facilitates arrestin-dependent Jun amino-terminal kinases (JNK) mitogen-activated protein kinase (MAPK) signaling. Mutation in the gene is associated with the risk to develop opiate addiction.
