一般描述

胞内蛋白酶 Glu-C 测序级是从 金黄色葡萄球菌 V8 中分离得到的丝氨酸蛋白酶,为高度纯化的特异性蛋白酶。胞内蛋白酶 Glu-C 的抑制剂有 DFP、α2-巨球蛋白和 TLCK。

特异性

特异性水解 Glu(碳酸氢铵,pH 7.8 或乙酸铵缓冲液,pH 4.0)或 Glu 和 Asp(磷酸盐缓冲液,pH 7.8)羧基侧肽键的丝氨酸蛋白酶。以胰岛素(胰岛素 B ox )的氧化 B 链为底物,确证了胞内蛋白酶 Glu-C 测序级的特异性。孵育高浓度的胞内蛋白酶 Glu-C 测序级(1 份(按重量计)酶和 10 份(按重量计)胰岛素 B ox ),以检测痕量杂质,例如污染蛋白酶。
热灭活: 大多数测序级蛋白酶通过煮沸 5 min 停止。

应用

使用胞内蛋白酶 Glu-C 测序级进行蛋白质结构分析和序列分析。[1][2]

特点和优势

不含可能干扰 RP HPLC 肽分离的杂质。

制备说明

工作浓度: 1/100 至 1/20 (w/w)
最小量 1~5μg 胰岛素 B ox,使用 Column Aquapore RUP 300,4.6 x 100 nm,7μm 可检测到足够的水平。
工作液: 推荐用双蒸水配制溶剂。
除水外,未检测其他溶剂;罗氏推荐使用 50 mM Hepes,pH 7.8。
储存条件(工作液): + 2 至 + 8°C 下储存 1-2 天。
稳定性: + 2 至 + 8°C 下储存 12 个月,在质量标准范围内;干燥储存。

重悬

冻干内蛋白酶 Glu-C 测序级在双蒸馏水中重构。 为避免自溶,培养温度不应超过 25°C。
