General description

MD-1 is a lymphocyte antigen 86 (LY86). The mature peptide is a secretory molecule, has 143 amino acids with no hydrophobic stretches. It has two N-terminal glycosylation sites. Human Md-1 is homologous to mice and chicken Md-1. Out of seven cysteine residues, six is conserved in all three species. MD-1 protein is expressed on the cell surface and lacks transmembrane region. MD-1/Radioprotective 105(RP105) complex is expressed on cell surfaces of B-cells, dendritic cells and macrophages. In human chromosome, the gene MD-1 is localised on 6p25.1.

Immunogen

a 15 amino acid peptide from near the center of human MD-1.

Biochem/physiol Actions

MD-1 associates with Radioprotective 105 (RP105) protein. MD-1 mediates cell surface expression of RP105. Together MD-1 and RP105 are required for lipopolysaccharide (LPS) recognition and signalling. Lack of MD-1 leads to reduced responsiveness to microbial membrane/LPS in B cells. Expression of MD-1 in dendritic cells and macrophages leads to inhibition of TLR4 (Toll like receptor 4)/MD-2 responsiveness to LPS. Methylation of LY86 gene causes obesity and plays a key role in obesity related metabolic disorder.
