General description
CASK (Calcium/calmodulin-dependent serine protein kinase) interacting protein 1 (CASKIN1) is expressed in the neuronal cells. The N terminal region of Caskin1 have six ankyrin repeats, two sterile α motifs (SAM domains) and a single SH3 domain. The C-terminal region possesses a long proline-rich region. In human chromosome, the gene CASKIN1 is localized on 16p13.3.
Biochem/physiol Actions
CASK interacting protein 1 (CASKIN1) is a scaffold protein involved in synapses. Caskin1 interacts with Calcium/calmodulin-dependent serine protein kinase (Cask) in the CASK interaction domain (CID) adjacent to SAM domain. Caskin1 also interacts with Nck (non-catalytic region of tyrosine kinase adaptor protein 1)/Dock (dedicator of cytokinesis), neurexin2, synaptotagmin, and septin4. Caskin1 interacts with EphB1 (ephrinB ligands) through Nck adapter protein. The interaction phosphorylates the tyrosine residue in the SH3 domain and causes changes in the SH3 domain. Caskin1 forms a tripartite complex with Cask/caskin1/velis. Caskin1 competes with Munc-interacting protein 1 (Mint1) to form the tripartite complex. The tripartite complex binds to the cytoplasmic tail of neurexin, which is enriched in the presynaptic sites.
