Preparation Note

Application

胰蛋白酶-EDTA溶液已用于：
  • 在高浓度样品的相对细胞频率测定期间，对补充10%胎牛血清的RPMI 1640培养的HT29人结直肠癌细胞进行分离。
  • 胰蛋白酶消化瞬时转染的人胚肾tcA-201细胞系。
  • 在细胞培养期间酶促释放粘附于支架的小鼠成纤维细胞（L929细胞系），以评估几种聚(L/D)丙交酯96/4无纺布支架和纤维改性处理的影响。
  • 分离培养皿中的细胞进行流式细胞术分析。
适合用于制备测序用单细胞悬液。
该产品的常见用途在于将贴壁细胞从培养基表面移除。 将细胞从基底上分离所需的胰蛋白酶浓度主要依赖于细胞类型和培养基已使用的时间。

Biochem/physiol Actions

胰蛋白酶可切割赖氨酸和精氨酸残基的C末端肽段。 如果酸性残基位于切割位点的任意一侧，则该反应的水解速度减慢，并且如果脯氨酸残基位于切割位点的羧基侧，则水解停止。 胰蛋白酶活性的最佳pH条件为7-9。 胰蛋白酶还可以切割氨基酸合成衍生物的酯和酰胺键。 将EDTA添加到胰蛋白酶溶液中作为螯合剂，用于中和钙离子和镁离子以防止其抑制胰蛋白酶活性。 去除这些离子会增加酶活性。

丝氨酸蛋白酶抑制剂（包括DFP、TLCK、APMSF、AEBSEF和抑肽酶等）会抑制胰蛋白酶。

General description

胰蛋白酶由含有223个氨基酸残基的单链多肽组成，由胰蛋白酶原在Lys - lle肽键处切去N-端六肽而得。 这些氨基酸序列通过6个二硫键交联。 这就是胰蛋白酶（即β-胰蛋白酶）的天然形式。 BETA-胰蛋白酶可自溶，在Lys-Ser残基裂解，产生α-胰蛋白酶。 胰蛋白酶是丝氨酸蛋白酶家族的成员。
