General description

A Certified Reference Material of the major urinary metabolite of methadone. Methadone, sold under various trade names such as Symoron, Amidone or Methadose®, is a synthetic opioid used medically as an analgesic and as a maintenance anti-addictive for use in patients with opioid dependency. Suitable uses for this reference standard include as starting material in preparation of calibrators and controls for LC/MS or GC/MS methods in methadone testing for applications in clinical toxicology or forensic analysis.

We developed a method based on liquid chromatography coupled with tandem mass spectrometry to quantify individual enantiomers of methadone and its primary metabolite, R/S-2-ethyl-1,5-dimethyl-3,3-diphenylpyrrolinium (EDDP), in postmortem blood and brain tissue. Samples were prepared with a Tecan Evo robotic system.

Methadone maintenance therapy is one of the most popular treatments for opiate addiction. The easiest and the most reliable way to monitor the accuracy of the therapy is determination of methadone and its metabolites in biological material. The aim of

Body fluids and tissues in 16 methadone (MTD)-associated fatalities were investigated to find out whether analysis of MTD and its metabolite 2-ethylidine-1,5-dimethyl-3,3-diphenylpyrrolidine (EDDP) following enantioselective separation of both compounds may assist in the interpretation of MTD findings. Individual case histories

Methadone is used to treat moderate to severe pain in patients not responsive to non-narcotic analgesics and for maintenance treatment of opioid addiction. Methadone is primarily metabolized by N-demethylation to an inactive metabolite 2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidene (EDDP) by CYP3A4 and CYP2B6. Establishing
