General description

Programmed cell death protein 1 (PD-1), CD279, is a type I membrane protein found in T cells, B cells, and macrophages that binds Programmed cell death ligand 1 and 2 (PD-L1 and PD-L2). PD-1 is found in both soluble and insoluble forms. PD-1 regulates the body’s immune response through a variety of pathways, down-regulating immune system activity and suppressing T-cell inflammatory activity. PD-1 has been implicated as playing a role in cancers, autoimmune diseases, the immune response to viruses, and in immunological pathways related to brain repair. As an immune checkpoint, PD-1 promotes apoptosis of antigen-specific T-cells in lymph nodes and reduces apoptosis in regulatory T-cells. Blockers of the PD-1 pathway have been approved as cancer therapeutics.

Application

Immuno-Oncology

Features and Benefits

  • Next-Gen Sensitivity For Low-Expression Targets: Designed to quantify PD-1 even in samples with minimal immune checkpoint activity.
  • Illuminate Immune Exhaustion: Quantify PD-1 expression on CD8+ T cells to study T-cell exhaustion in cancer and chronic infections.
  • Advance Immuno-Oncology Research: Track soluble PD-1 as a biomarker of immune suppression, tumor progression, or therapeutic response.
  • Support Autoimmune and Viral Studies: Investigate PD-1’s role in immune regulation, IL-10 induction, and CD4+ T-cell inhibition.

Analysis Note

  • Sensitivity: LLOQ 3,470 fg/mL* (SMCxPRO® Instrument)
  • Standard Curve Range: 870-3,000,000 fg/mL
  • Intra-assay CV: <15%
  • Inter-assay CV: <20%
  • Linearity: Serum: 107% Plasma: 83%
  • Recovery: Serum: 93% Plasma 83%
*Indicative LLOQ created from average of 14 independent in house runs, where >80% met same value defined with CV & Recovery +/- 20%. Refer to CoA for lot specific performance.

Other Notes

Although PD-1 was detectable in rat, mouse, monkey, and canine samples, the assay has not been verified for these species.
