General description

Programmed cell death protein ligand 1 (PD-L1), also known as cluster of differentiation 274 (CD274) and B7-H1, is a type 1 membrane protein of 290 amino acids. It is found in a variety of cell types and binds programmed cell death protein (PD-1). It has been indicated as playing a role in autoimmune disease, allograft responses, pregnancy, and other disease states. When PD-L1 binds to PD-1, an inhibitory signal is transmitted that ultimately reduces the proliferation of antigen-specific T cells and reduces the rate of apoptosis in regulatory T-cells. By doing so, PD-L1 contributes to the immune evasion of cancer tissues by suppressing the adaptive immune system. Blocking this activity is thought to restore anti-tumor activity. High PD-L1 expression is correlated with poor prognosis with some cancer types. High levels of the soluble form are also associated with poor prognosis and advanced tumor progression. Multiple PD-L1 inhibiting monoclonal antibodies have been approved as cancer therapeutics.

Application

Immuno-Oncology

Features and Benefits

  • Unlock Hidden Insights in Cancer Immunology: Detect soluble PD-L1 levels associated with tumor progression and poor prognosis, even at sub-picogram concentrations.
  • Accelerate Biomarker Discovery: Ideal for studying immune checkpoint dynamics in cancer, autoimmune disease, and transplant biology.
  • Confidence in Every Data Point: High recovery rates and linearity ensure data integrity across sample types. SMC® T
  • Technology: Digitally detects PD-L1 at ultrasensitive levels—down to 780 fg/mL, ideal for low-expression biomarker studies.

Analysis Note

  • Sensitivity: LLOQ 780 fg/mL* (SMCxPRO® Instrument)
  • Standard Curve Range: 390-400,000 fg/mL
  • Intra-assay CV: <15%
  • Inter-assay CV: <20%
  • Linearity: Serum: 120% Plasma: 110%
  • Recovery: Serum: 117% Plasma: 109%
*Indicative LLOQ created from average of 14 independent in house runs, where >80% met same value defined with CV & Recovery +/- 20%. Refer to CoA for lot specific performance.

Other Notes

Although PD-L1 was detectable in rat, mouse, monkey, and canine samples, the assay has not been verified for these species.
