General description

GTP-binding proteins (G-proteins) play a critical role in signal transduction by coupling receptors to effectors. The intracellular consequences of G-protein activation include second messenger generation, protein phosphorylation, ion channel activation, gene induction, cell growth, and differentiation. Gi belongs to the pertussis toxin sensitive G-proteins. Gi consists of three different subtypes of alpha-subunits (alphai1, alphai2 and alphai3).
~40 kDa

Immunogen

Peptide corresponding to amino acids 345-354 of Gi1 and 346-355 of Gi2

Application

Anti-G Protein Giα 1/2 Antibody is an antibody against G Protein Giα 1/2 for use in WB, IH(P).
Immunohistochemistry(paraffin):
Representative testing from a previous lot.
Optimal Staining of GI ALPHA 1/2 With Citrate pH 6.0 Epitope Retrieval: Rat Cerebellum GI ALPHA 1/2 representative staining pattern/morphology in rat cerebellum. Tissue was pretreated with citrate pH 6, antigen retrieval. A previous lot of this antibody was diluted to 1:100, using IHC-Select Detection with HRP-DAB.

Optimal working dilutions must be determined by end user.

Biochem/physiol Actions

Recognizes G protein subunit αi1.

Physical form

Format: Purified

Preparation Note

Stable at -20°C in undiluted aliquots for up to 12 months from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Routinely evaluated by Western Blot on L6 lysates.

Western Blot Analysis: 1:500 dilution of this lot detected G protein Gialpha 1/2 on 10 μg of L6 lysates.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.
