General description

A cell-permeable, arylsulphonamidyl thiophene amide that is shown to reduce IL-6-induced STAT3 transcriptional activity (EC50 = 15 µM) in a luciferase reporter assay, in HeLa cells. It selectively inhibits STAT3, but not STAT1, phosphorylation following IL-6 stimulation in both HeLa and MDA-MB-231 breast cancer cells at concentrations of 10-30 &micro:M and 30-100 µM, respectively. Following IL-6 stimulation, it suppresses the cell viability of STAT3-dependent cancer cells by 40% compared with that of STAT3-null cancer cells at the low micromolar level.
