General description
PRONASE® Protease, Streptomyces griseus, nuclease-free is a non-specific protease that liquefies mucins and digests proteins to free amino acids. This mixture can be used to isolate living chondrocytes. It is most active at pH 7.0 but stable over wide ranges of pH and temperature. It has an optimal pH of 7.5. Pronase is less cytotoxic and produces more efficient dissociation than collagenase.
Application
Pronase® Protease, Streptomyces griseus, nuclease-free has been used:
- in single-cell preparations for cardiac side population (CSP) analysis
- in aflatoxin plasma biomarker assay
- to permeabilize Caenorhabditis elegans embryo for in vivo micrococcal nuclease digestion
Biochem/physiol Actions
Proteases catalyze the hydrolysis of proteins to form peptides and free amino acids. They also regulate the breakdown of fats and carbohydrates through many enzymatic cascades and activating proenzyme zymogens.
Preparation Note
Unstable in solution; reconstitute just prior to use.
Other Notes
Note: 1 KU = 1000 units.
One unit is defined as the amount of enzyme that will liberate a digestion product equivalent to 25 µg of tyrosine per min at 40°C, pH 7.5.
