pCOLADuet-1 DNA - Novagen^®

pCOLADuet-1设计用于从单个质粒共表达两个靶基因。该载体编码两个多克隆位点(MCS)，每个位点之前都有一个T7启动子、lac操纵子和核糖体结合位点(rbs)。如果需要，MCS-1编码用于创建N端融合的六个氨基酸的His•Tag序列，而MCS2编码在最后一个限制性位点之后用于创建C端融合的15个氨基酸的S•Tag肽。可以使用ACYCDuetUP1引物和DuetDOWN1引物对插入MCS-1的基因进行测序。可以使用DuetUP2引物和T7终止子引物对插入MCS-2的基因进行测序。该载体具有来自ColA(1)的COLA复制子和卡那霉素抗性基因。可以用含有兼容的复制起点和耐药基因的质粒将该载体转化到同一细胞中，以共表达多达八个靶基因。
