Immunogen

Highly purified peptide corresponding to residues 254-271 of rat or mouse Aquaporin 2 (SwissProt accession number P34080).

Application

Anti-Aquaporin 2 Antibody is an antibody against Aquaporin 2 for use in WB, IH.
Research Category
Neuroscience
Research Sub Category
Ion Channels & Transporters
Western blotting: 1:200-1:1000 using ECL on rat kidney membranes.

Immunohistochemistry on rat kidney sections.

Dilutions should be made using a carrier protein such as BSA (1-3%).

Optimal working dilutions must be determined by the end user.

Biochem/physiol Actions

Recognizes Aquaporin 2.

SPECIES REACTIVITIES: The epitope specific for Aquaporin 2 is identical in rat and mouse and only slightly different in human (17/18 amino acids). Reactivity with other species has not yet been tested.

Physical form

Affinity purified immunoglobulin. Lyophilized from PBS, pH 7.4, containing 1% BSA and 0.05% sodium azide. Reconstitute with 200 μL of sterile distilled water. Centrifuge antibody preparation before use (10,000 x g for 5 min).

Preparation Note

Maintain lyophilized material at -20°C for up to 6 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Analysis Note

Control
Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). Reconstitute with 100 μL of deionized water. For negative control, preincubate 0.5ug of purified peptide with 1 μg of antibody for one hour at room temperature.

Optimal concentrations must be determined by the end user.

Other Notes

Concentration: Please refer to the Certificate of Analysis for the lot-specific concentration.

Arginine vasopressin (AVP) mediates water reabsorption in the kidney collecting ducts through regulation of aquaporin-2 (AQP2). Also, estrogen has been known to regulate AQP2. Consistently, we previously demonstrated that tamoxifen (TAM), a selective estrogen receptor modulator, attenuates the downregulation of

Because cyclophosphamide-induced hyponatremia was reported to occur without changes in plasma vasopressin in a patient with central diabetes insipidus, we hypothesized that cyclophosphamide or its active metabolite, 4-hydroperoxycyclophosphamide (4-HC), may directly dysregulate the expression of water channels or sodium transporters

However, ammonia-specific transporters, Mep/Amt proteins, are present in primitive organisms and mammals express orthologs of Mep/Amt proteins, the Rh glycoproteins. These findings suggest that the mechanisms of
