General description
FUNCTION: SwissProt: O95279 # pH-dependent, voltage insensitive, outwardly rectifying potassium channel. Outward rectification is lost at high external K(+) concentrations.
SIZE: 499 amino acids; 55130 Da
SUBUNIT: Homodimer; disulfide-linked.
SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein (Potential).
TISSUE SPECIFICITY: Abundant expression in kidney, also detected in liver, placenta and small intestine. In the kidney, expression is restricted to the distal tubules and collecting ducts. Not expressed in proximal tubules or glomeruli.
SIMILARITY: SwissProt: O95279 ## Belongs to the two pore domain potassium channel (TC 1.A.1.8) family.
MISCELLANEOUS: Inhibited by quinine, quinidine and external acidification.

MISCELLANEOUS: Inhibited by quinine, quinidine and external acidification.
A purified peptide (TASK-2) corresponding to amino acid residues 483-499 of human TASK-2 (Accession O95279).
Application
Western blot: 1.5 μg/mL (1:200) on rat kidney membranes using ECL.
Immunohistochemistry on rat kidney tissue sections.
Dilutions should be made using a carrier protein such as BSA (1-3%).
Optimal working dilutions must be determined by the end user.

Biochem/physiol Actions
The epitope is specific for TASK-2 and is not present in any other known proteins.
Physical form
Affinity purified immunoglobulin. Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, 5% sucrose as a stabilizer and 0.025% sodium azide as a preservative. Reconstitute with 50 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).
Preparation Note
Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.
Other Notes
CONTROL ANTIGEN: Included free of charge with the antibody is 20 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of purified peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
