FUNCTION: SwissProt: Q9NSA2 # Pore-forming (alpha) subunit of voltage-gated rapidly inactivating A-type potassium channels. May contribute to I(To) current in heart and I(Sa) current in neurons. Channel properties are modulated by interactions with other alpha subunits and with regulatory subunits.

SUBCELLULAR LOCATION: Membrane; Multi-pass membrane protein.
TISSUE SPECIFICITY: Widely expressed. Highly expressed in brain, in particular in cerebellum and thalamus; detected at lower levels in the other parts of the brain.
DOMAIN:SwissProt: Q9NSA2 The segment S4 is probably the voltage-sensor and is characterized by a series of positively charged amino acids at every third position.
SIMILARITY: Belongs to the potassium channel family. D (Shal) subfamily.

Purified peptide from amino acids 538-550 of human K_v4.1

Western blot: 1:400 using ECL on rat and mouse brain lysate and human SH-SY5Y neuroblastoma cell line.

Dilutions should be made using a carrier protein such as BSA (1-3%).

Optimal working dilutions must be determined by the end user.

Recognizes Kv4.1 (Voltage-gated K+ channel subfamily D member 1, KCND1).

Lyophilized from phosphate buffered saline, pH 7.4, containing 1% BSA, and 0.025% sodium azide as a preservative. Reconstitute with 200 μL of sterile deionized water. Centrifuge antibody preparation before use (10,000 xg for 5 min).

Maintain lyophilized material at -20°C for up to 12 months after date of receipt. After reconstitution maintain at -20°C in undiluted aliquots for up to 6 months. Avoid repeated freeze/thaw cycles.

Included free of charge with the antibody is 40 μg of control antigen (lyophilized powder). The stock solution of the antigen can be made up using 100 μL of sterile deionized water. For negative control, preincubate 1 μg of peptide with 1 μg of antibody for one hour at room temperature. Optimal concentrations must be determined by the end user.
