General description
Also known as ARD1 (Activator of RNA decay), NIPP-1 (nuclear inhibitor of protein phosphatase 1) is a ubiquitously expressed multifunctional nuclear scaffold protein of 39 kDa, which plays an important role in integrating transcription, RNA processing, and cell cycle progression. NIPP-1 serves as a signaling hub in the nucleus. Additionally, NIPP-1 contains at least two binding sites for PP1 and is complexed to about one-third of the nuclear pool of PP1. Normally, the NIPP-1 - PP1 complex is inactive, but can be activated following phosphorylation of NIPP-1 with PKA, CFK2, or Src family kinases. NIPP-1 also interacts directly with nucleic acids and components of polycomb repressive complexe 2 (PRC2) and functions as a PRC2-dependent transcriptional repressor.
~42 kDa observed. Three isoforms at 39 kDa (alpha isoform), 23 kDa (beta isoform), and 13 kDa (gamma isoform) are known to exist. An uncharacterized band may be observed at ~62 kDa in some nuclear extracts.
