General description
Inactive serine/threonine-protein kinase TEX14 (UniProt: Q8IWB6; also known as Protein kinase-like protein SgK307, Sugen kinase 307, Testis-expressed sequence 14, Testis-expressed sequence 14 protein) is encoded by the TEX14 (also known as SGK307) gene (Gene ID: 56155) in human. TEX14 is a testis expressed, Plk-1 regulated protein that is required both for the formation of intercellular bridges during meiosis and for kinetochore-microtubule attachment during mitosis. Intercellular bridges connect differentiating germ cells and are required for spermatogenesis and male fertility. Three isoforms of TEX14 have been described that are produced by alternative splicing. TEX14 promotes the conversion of midbodies into intercellular bridges via its interaction with CEP55. Its interaction with CEP55 inhibits the interaction between CEP55 and PDCD6IP/ALIX and TSG101, which blocks cell abscission and transforms midbodies into intercellular bridges. TEX14 has three ANK domains (aa 27-54; 55-84, and 88-117) and a protein kinase domain. The protein kinase domain is catalytically inactive. TEX14 is phosphorylated on threonine residues by CDK1 during early phases of mitosis, which promotes the interaction with PLK1 and recruitment to kinetochores. It is also phosphorylated on Ser437 by PLK1 during late prometaphase, which promotes the rapid depletion from kinetochores and its subsequent degradation by the APC/C complex. Immunohistochemistry Analysis: A 1:250-1,000 dilution from a representative lot detected TEX14 in human testis and human ovary tissues.
