General description
Cyclic AMP-dependent transcription factor ATF-5 (UniProt: also known as cAMP-dependent transcription factor ATF-5, Activating transcription factor 5, Transcription factor ATFx) is encoded by the ATF5 (also known as ATFX) gene (Gene ID: 22809) in human. ATF5 belongs to the ATF/CREB family of transcription factors that is widely expressed. It has been implicated in multiple biological processes, such as regulation of cell proliferation and differentiation, cell stress response and survival. ATF5 can either stimulate or repress gene transcription through binding of different DNA regulatory elements such as CRE (cAMP response element), ARE (ATF5 regulatory element), and AARE (amino acid response element). Its transcriptional activity is enhanced by CCND3 and slightly inhibited by CDK4. ATF5 is an essential structural protein that is required for the interaction between the mother centriole and the pericentriolar material (PCM). ATF5 depletion is reported to block PCM accumulation at the centrosome and cause fragmentation of centrioles, which contributes to the formation of multi-polar mitotic spindles and genomic instability. ATF5 homeostasis is essential for the function of the centrosome and for the global microtubule organization.
