Gene Information
mouse ... Nrxn1(18189)
General description
Neurexin-1 (UniProt Q9CS84; also known as Neurexin I-alpha, Neurexin-1-alpha) is encoded by the Nrxn1 (also known as Kiaa0578) gene (Gene ID 18189) in murine species. Neurexin (NRXN) is a presynaptic single pass transmembrane protein that assists in joining neurons together at the synapse. Neurexins mediate signaling across the synapse and neural networks by specifying synpatic function. Mutations in the genes encoding for neurexins are implicated in autism, schizphrenia, and nicotine dependence. There are two classifications of neurexins, α-NRXNs and β-NRXNs. The α-NRXNs are larger and have different amino-terminal extracellular sequences than β-NRXNs. β-NRXNs act as receptors for neuroligin and have been found to play a role in angiogenesis.
~170/55/40 kDa observed. Varible depending on the isoform(s) present in the test sample(s).
Immunogen
Epitope: Near C-terminus.
KLH-conjugated linear peptide corresponding to a cytoplasmic domain sequence near the C-teminus of mouse neurexin-1.
Application
Anti-pan Neurexin-1 Antibody detects level of pan Neurexin-1 and has been published and validated for use in Western Blotting, Immunohistochemistry (Paraffin).
Immunohistochemistry Analysis: A 1:1,000 dilution from a representative lot detected Neurexin-1 in mouse, rat, and human cerebral cortex tissue sections.

Biochem/physiol Actions
Expected to react with all neurexin-1-alpha and neurexin-1-beta spliced isoforms of murine, human, and rat species, as this polyclonal antibody targets the C-terminal end cytoplasmic sequence that is not affected by alternative splicings (Missler, M., and Südhof, T.C. (1998). Trends Genet. 14(1):20-26). Cross-reactivities toward alpha and beta isoforms of neurexin-2 and neurexin-3 are also likely, although the C-termminal sequence is not 100% conserved among neurexin-1, neurexin-2 and neurexin-3.
Physical form
Affinity purified
Purified rabbit polyclonal antibody in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
Preparation Note
Stable for 1 year at 2-8°C from date of receipt.
Analysis Note
Evaluated by Western Blotting in mouse brain tissue lysate.
Western Blotting Analysis: 0.2 µg/mL of this antibody detected Neurexin-1 in 10 µg of mouse brain tissue lysate.
