General description

The Na-K-Cl cotransporter 1 (NKCC1) is a membrane protein coded by the SLC12A2 gene and functions in cellular sodium/potassium/chloride transport and homeostasis. It is widely distributed throughout the body and serves important functions, especially in liquid-secreting glands. Phosphorylation of Thr212/Thr217 in the intracellular N-terminal domain of NKCC1 is correlated with NKCC1 activation.
~250 kDa observed. Likely a dimer of mature form of NKCC1, NKCC1 is glycosylated differently in different cells, leading to uncharacterized band(s) in some lysates.

Preparation Note

Stable for 1 year at -20°C from date of receipt.
Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Analysis Note

Evaluated by Western Blotting in GMB43 cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected phospho-NKCC1 Thr212/Thr217 in 10 µg of GMB43 cell lysate.
