General description

The CHEMICON in vitro angiogenesis assay kit represents a simple model of angiogenesis in which the induction or inhibition of tube formation by exogenous signals can be easily monitored. Angiogenesis is the process of generating new capillary blood vessels. It is a fundamental component of several normal (reproduction and wound healing) and pathological processes (diabetic retinopathy, rheumatoid arthritis, tumor growth, and metastasis).
The CHEMICON in vitro angiogenesis assay kit provides a convenient system for evaluation of tube formation by endothelial cells in a convenient 96-well format. When cultured on ECMatrix, a solid gel of basement proteins prepared from the Engelbreth Holm-Swarm (EHS) mouse tumor, these endothelial cells rapidly align and form hollow tube-like structures. Tube formation is a multi-step process involving cell adhesion, migration, differentiation, and growth.
ECMatrix is optimized for maximal tube-formation. For assaying inhibitors or stimulators of tube formation, simply premix the endothelial cell suspension with different concentrations of the inhibitor or stimulator to be tested, before adding the cells to the top of the ECMatrix.

Application

In vitro angiogenesis assay kit has been used to monitor the extent of tube assembly/angiogenic potential in various endothelial cells such as:
  • human umbilical vein cells (HUVEC)
  • primary human retinal microvascular cells
  • platelet derived growth factor receptor-β (PDGFRβ)/PAE cells
  • bovine capillary endothelial (BCE) cells

Preparation Note

Store kit materials at -20 ºC for up to 6 months. Once ECMatrix has been thawed, store at 2-8 ºC for one week. Do not refreeze.

Other Notes

  • ECMatrixTM Gel Solution: (Part No. 90060) Five 1 mL bottles.
  • ECMatrixTM Diluent Buffer, 10X: (Part No. 90061) One 1 mL bottle.
The CHEMICON in vitro angiogenesis assay kit is intended for research use only; not for diagnostic or therapeutic applications.

"The successful, reliable culture of epithelial cells is critical for many areas of research, including dermatology, respiratory research, and cancer research. Because the breakdown of control mechanisms in epithelial cells is a frequent contributor to cancer progression and metastasis, epithelial cell culture is particularly important for cancer research. EpiGRO™ media formulations are optimized to provide better viability, proliferation rates, morphology and culture stability than other commercially available options. The media are provided in unique, light-blocking, temperature-monitored packaging to ensure stability and protect the media from damage by light, contamination, and excessive heat. The media do not require or contain any antimicrobials or phenol red. These components can cause cell stress and influence experimental results by masking the true performance or health of the cell culture. Phenol red acts like an estrogen and may stimulate growth independently of experimental variables. "

Key Steps in Tumor Metastasis

"Recognizing both the tremendous opportunities and the challenges facing cancer research, we are dedicated to developing and refining tools and technologies for the study of cancer. With our comprehensive portfolio, including the Upstate®, Chemicon®, and Calbiochem® brands of reagents and antibodies, researchers can count on dependable, high quality solutions for analyzing all the hallmarks of cancer."

Our team of scientists has experience in all areas of research including Life Science, Material Science, Chemical Synthesis, Chromatography, Analytical and many others.
