General description

Bovine myocardial m-calpain.

Specificity

Reacts with calpain small subunit.

Application

ELISA Analysis: 1:50,000 (against the immunogen).
Western Blotting Analysis: A representative lot detected steady level of cellular calpain small subunit during the final stage of human CD34+ erythroid progenitor cells differentiation to reticulocytes (Patterson, T., et al. (2009). J. Biol. Chem. 284(21):14547-1457).
Western Blotting Analysis: A representative lot detected upregulated calpain small subunit level in atrophied soleus muscle upon hind limbs immobilization by plaster cast among male rats (Sugiura, T., et al. (2003). J. Physiol. Sci. 56(6):393-399).
Western Blotting Analysis: A representative lot detected calpain small subunit in human WI-38 fibroblasts (Zhang, W., et al. (1996). J. Biol. Chem. 271(31):18825-18830).
Immunocytochemistry Analysis: A representative lot detected increrased plasma membrane calpain small subunit immunoreactivity following ionomcyin treatment of human large cell lung carcinoma LCLC-103H cells by fluorescent immunocytochemistry (Gil-Parrado, S., et al. (2003). J. Biol. Chem. 278(18):16336-16346).
Immunoprecipitation Analysis: Representative lots immunoprecipitated calpain small subunit from human WI-38 fibroblasts (Zhang, W., et al. (1996). J. Biol. Chem. 271(31):18825-18830; Zhang, W., and Mellgren, R.L. (1996). Biochem. Biophys. Res. Commun. 227(3):891-896.).
Activity Assay: A representative lot inhibited the proteolytic activity of bovine and canine calpain 2, as well as partially purified human erythrocyte calpain I as a result of increased Km and decreased Vmax toward the substrate casein (Mellgren, R.L., and Lane, R.D. (1988). Biochem. Biophys. Res. Commun. 954(2):154-160).

Quality

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected Calpain, small subunit in 10 µg of HEK293 cell lysate.

Physical properties

~30 kDa observed

Physical form

Purified mouse monoclonal IgG1κ antibody in PBS without preservatives.

Storage and Stability

Stable for 1 year at -20°C from date of receipt.Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Other Notes

Western Blotting Analysis: 0.5 µg/mL from a representative lot detected Calpain, small subunit in 10 µg of SHSY-5Y cell lysate.
Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected Calpain, small subunit in human kidney, human testis, and human prostate tissue.
