General description
Sorting nexin-17 (UniProt Q15036) is encoded by the SNX17 (also known as KIAA0064) gene (Gene ID 9784) in human. The sorting nexins (SNXs) constitute a family of adaptor proteins involved in proteins trafficking through the endosomal pathway. Originally identified as a binding partner of the cell adhesion protein P-selectin, sorting nexin-17 (SNX17) has since been shown to mediate internalization and recycling of multiple cell surface proteins including P-selectin, amyloid precursor protein (APP), integrin β1, low density lipoprotein receptor (LDLR), LDLR related protein (Lrp1), ApoER2, and FEEL1. All SNX proteins contain a conserved phox homology (PX) domain that mediates phosphoinositides binding in a SNX type-specific manner, as well as various scaffolding modules including SH3, BAR, and PDZ domains. SNX17 PX domain (a.a. 1-109) displays specificity for phosphatidylinositol 3-monophosphate, which is enriched at the cytoplasmic face of the early endosome. SNX17 also contains a 4.1 protein/ezrin/radixin/moesin (FERM) domain (a.a. 115-432) comprised of an ubiquitin/Ras association (RA) subdomain known as the F1 lobe (a.a. 113-205), an F2 lobe (a.a. 206-269) with a helical structure similar to acyl-CoA binding protein, and an F3 lobe (a.a. 270-388) similar to the phosphotyrosine binding (PTB)/pleckstrin homology domain. The FERM domain mediates interaction with Asn-Pro-Xaa-Tyr/Phe (NPX(Y/F) motif-containing proteins for internalization as well as interaction with adaptor proteins-containing PTB (or FERM) domains to trigger signaling events.
Specificity
Clone 4B8.1 recognizes an internal epitope within the FERM-like region present in both spliced isoforms of human sorting nexin-17 reported by UniProt (Q15036).
