Gene Information
human ... KLF4(9314)
General description
Krueppel-like factor 4 (KLF4), which is also known as epithelial zinc finger protein (EZF) and gut-enriched krueppel-like factor (GKLF), is a developmentally regulated transcription factor that plays an important role in maintaining the pluripotent state in embryonic stem cells (ESCs) and is used as a marker of the pluripotent state for cells in culture. It is required for the differentiation of epithelial cells and the establishment of the barrier function of the skin, and KLF4 is also expressed during skeletal and kidney development. KLF4 is highly expressed in the skin, lung, and intestine along the epithelial border. This nuclear localized protein can act as both a tumor suppressor and activator depending upon its environment, but its most prominent function is growth suppression. Interestingly, increased expression of KLF4 leads to an increase in expression of P21WAF1/CIP1 which is an important checkpoint protein that inhibits cell cycle progression and is essential in mediating the cell cycle arrest at both the G1/S and G2/M boundaries. Additionally, KLF4 mediates the normal functions of the tumor suppressor p53 as well. Mutations thus in KLF4 lead to unchecked growth and tumor formation, and because of its expression and involvement within the gut, KLF4 is often mutated in many colon cancer cell lines and the lack of KLF4 expression is correlated with colorectal cancer pathogenesis. Elevated expression of KLF4 gene expression has also been associated with gastric cancer, breast cancer, and squamous cell carcinomas. EMD-Millipore’s Anti-KLF4 mouse monoclonal antibody, clone 1E6, has been tested in western blot against recombinant hKLF4 expressed cell lysates and in immunocytochemistry on paraffin embedded human lung and colon cancer tissue. The clone has also been tested in fluorescent immunocytochemistry on the human esophageal squamous carcinoma cell line Eca109 in culture.
~55 kDa observed. Uncharacterized bands may appear in some lysate(s). Uniprot describes 3 isoforms between ~46 kDa and ~55 kDa
Immunogen
Purified recombinant fragment of human KLF4 expressed in E. coli.
Application
Detect KLF4 using this mouse monoclonal antibody, Anti-KLF4 Antibody, clone 1E6 validated for use in IHC & western blotting.
Immunofluorescence Analysis: A 1:200-1,000 dilution from a representative lot detected KLF4 in ECA109 cells.
Western Blotting Analysis: A 1:3,000 dilution from a representative lot detected KLF4 in transfected HEK293 cell lysates.
Optimal working dilutions must be determined by end user.
