General description

Lysine-specific demethylase 3A (UniProt: Q6PCM1; also known as EC:1.14.11.65, JmjC domain-containing histone demethylation protein 2A, Jumonji domain-containing protein 1A, [histone H3]-dimethyl-L-lysine(9) demethylase 3A) is encoded by the Kdm3a (also known as Jhdm2a, Jmjd1a, Kiaa0742) gene (Gene ID: 104263) in mouse. KDM3A is a member of the Jumonji C (JmjC) domain-containing family of histone demethylases. KDM3A contains a C-terminal JmjC domain that catalyzes the demethylation of mono- and di-methylated lysine 9 on histone H3 (H3K9me1/me2), a modification associated with transcriptional repression. In addition, it contains a zinc-finger domain for DNA binding and an LXXLL motif for interacting with nuclear receptors. As a result of KDM3A demethylation of H3K9me1/me2, gene transcription increases. This enzymatic activity plays a crucial role in regulating gene expression by altering chromatin structure, thereby influencing various biological processes such as cell differentiation, proliferation, spermatogenesis, energy metabolism, stem cell regulation, and development. JMJD1A is also involved in beige adipogenesis through a multistep process where phosphorylation of S265 of JMJD1A stimulates its H3K9me2 demethylase activity that ultimately results in the thermogenic gene regulation in beige adipogenesis. KDM3A is expressed in a wide range of tissues, with particularly high levels in the testes, reflecting its role in spermatogenesis. The involvement of KDM3A in disease has been increasingly recognized, particularly in cancer, where it is often overexpressed and contributes to tumor progression by activating oncogenes and promoting cell cycle progression. Additionally, KDM3A has been implicated in metabolic disorders and cardiovascular diseases. The therapeutic potential of targeting KDM3A in disease contexts is an area of active research.
