General description
DBF4, also known as Protein DBF4 homolog A, Activator of S phase kinase, also known as Chiffon homolog A or DBF4-type zinc finger containing protein 1, and encoded by the gene DBF4/ASK/DBF4A/ZDBF1, is the regulatory subunit for the cell cycle kinase CDC7. The Cdc7 kinase is involved in regulation of the cell cycle at the point of chromosomal DNA replication. The complex CDC7-DBF4A selectively phosphorylates MCM2 subunit at ′Ser-40′ and ′Ser-53′ and then is involved in regulating the initiation of DNA replication during cell cycle. . DBF4 Forms a complex with CDC7. Note that CDC7 forms distinct complex either with DBF4A or DBF4B. Such complexes are stable upon replication stress. Interacts with MEN1, MCM2, ORC2, ORC4 and ORC6 proteins. DBF4 is localized to the nucleus. DBF4 is expressed in expressed in many cells particularly high in testis, thymus and most cancer cells.
