Natural cytotoxicity triggering receptor 1 (UniProt: S4S7F8; also known as Lymphocyte antigen 94 homolog, NK cell-activating receptor, Natural killer cell p46-related protein, NK-p46, NKp46, CD335) is encoded by the NCR1 gene (Gene ID: 611390) in canine species. NK-p46 is a single-pass type I membrane protein that belongs to the natural cytotoxicity receptor (NCR) family and serves as a cytotoxicity-activating receptor that may contribute to the increased efficiency of activated natural killer (NK) cells to mediate tumor cell lysis and elimination of virus-infected cells. It is considered as a primary activating receptor mediating NK cell antiviral function. It is involved in the functional recognition of viral HA and hemagglutinin-neuraminidases. Following the initiation of an NK-target cell interaction, NKp46 is reported to cluster at the cell membrane, specifically at the immune synapse where it mediates cytoskeletal rearrangement and cellular polarization. Canine NK-p46 is synthesized with a signal peptide of 20 amino acids, which is subsequently cleaved off. The mature form has an extracellular domain (aa 21-255), a helical domain (aa 256-279), and a cytoplasmic domain (aa 280-316). It also has an Ig domain (129-211). Clone 48A is shown to recognize CD3 /NKp46+ cells that display phenotypic and functional similarities to human NK cells. Also, it recognizes a subset of T cells that are NKp46 positive.
