General description
HLA class II histocompatibility antigen, DQ alpha 1 chain (UniProt: P01909; also known as DC-1 alpha chain, DC-alpha, HLA-DCA, MHC class II DQA1) is encoded by the HLA-DQA1 gene (Gene ID: 3117) in human. HLA‑DQA1, a member of the MHC class II family, is a single-pass type I membrane glycoprotein, which is synthesized with a signal peptide (aa 1-23) that is subsequently cleaved off to produce the mature protein that contains an extracellular domain (aa 24-216), a transmembrane domain (aa 217-239), and a cytoplasmic domain (aa 240-254). It is also detected in endoplasmic reticulum (ER), Golgi apparatus, and endosome membranes. It is a heterodimer of an and a β subunit. It is reported to bind peptides derived from antigens that access the endocytic route of antigen presenting cells (APC) and presents them on the cell surface for recognition by the CD4 T-cells. Its peptide binding cleft accommodates peptides of 10-30 residues. Exogenous antigens that have been endocytosed by the APC are thus readily available for presentation via MHC II molecules. Autophagy is also shown to be a source of endogenous peptides and autophagosomes are reported to constitutively fuse with MHC class II loading compartments. Clone SPV-L3 is shown to recognize a class II MHC products different from HLA-DR. It is reported to block the cytotoxic activity of CTL clones JR-2-19 and JR-2-26. (Ref.: Spits, H., et al. (1984). Preparation Note
Store at -10°C to -25°C. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
