Quality Control Testing

Evaluated by Western Blotting with recombinant -synuclein phosphorylated S129 and recombinant -synuclein monomers.

Western Blotting Analysis (WB): A 1:1,000 dilution of this antibody detected -synuclein phosphorylated on serine 129 in recombinant -synuclein phosphorylated S129 protein, but not in recombinant -synuclein monomers.

General description

Alpha-synuclein (UniProt: P37840; also known as Non-A beta component of AD amyloid, Non-A4 component of amyloid precursor, NACP) is encoded by the SNCA (also known as NACP, PARK1) gene (Gene ID: 6622) in human. Alpha-synuclein is a membrane-bound protein in dopaminergic neurons that is involved in the regulation of dopamine release and transport. It is also reported to induce fibrillization of microtubule-associated protein tau and reduces neuronal responsiveness to various apoptotic stimuli. It is primarily expressed in the brain and is associated with the regulation of neurotransmitter release at synapses. Alpha-synuclein is also found in the cytoplasm, where it exists in a dynamic equilibrium between a soluble cytosolic form and a membrane-bound form. It associates with lipid membranes, particularly those of synaptic vesicles. Aggregation of alpha-synuclein is known to be a causative factor in the development of Parkinson′s disease (PD), which leads to the degeneration of dopaminergic neurons in the substantia nigra (SN) in the midbrain region. -synuclein can be phosphorylated, predominantly on serine residues by casein kinase 1 (CK1). Phosphorylation by CK1 appears to occur on residue (serine 129) distinct from the residue (tyrosine 125) phosphorylated by other kinase FYN. The phosphorylation of -synuclein at serine 129 (pS129) has been extensively studied due to its significant implications in neurodegenerative diseases, particularly Parkinson′s disease (PD) and other synucleinopathies. Phosphorylation of Ser129 is reported to be selective and extensive in synucleinopathy lesions and promotes insoluble fibril formation. This serves as a potential biomarker for disease diagnosis and progression and offers a promising target for therapeutic intervention. (Ref.: Vaikath, N.N., et al. (2015). Immunogen

KLH-conjugated linear peptide corresponding to 9 amino acids surrounding phosphoserine 129 from the C-terminal region of human -synuclein.
