General description

Cholesteryl ester transfer protein (UniProt P22687; also known as CETP, Lipid transfer protein I, LTP) is encoded by the CETP (also known as HDLCQ10) gene (Gene ID 1071) in Oryctolagus cuniculus (Rabbit) species. Cholesteryl ester transfer protein (CETP) or lipid transfer protein (LTP) mediates the transport/exhanges of triglycerides and cholesteryl esters (CEs) between very-low-density lipoproteins (VLDL)/low-density lipoproteins (LDL) and high-density lipoproteins (HDL) in both directions. CETP plays an important role in the reverse cholesterol transport (RCT) pathway that mediates the transport of cholesterol from peripheral cells and tissues to the liver for metabolism and excretion in the bile. CETP shuttles CEs from HDL particles to apolipoprotein B-/apoB-containing particles in exchange for triglycerides. CETP is thought to facilitate the generation of small, dense LDL (sd LDL) particles through an indirect mechanism of increased rate of triglyceride transfer from VLDL in exchange for CEs in LDL and HDL. CE transfer between lipoproteins plays a crucial role in intravascular LDL remodeling. Decreased levels of HDL cholesterol and elevated levels of LDL is a risk factor for atherosclerosis and coronary heart disease (CHD).

Specificity

Clone 14-8F (a.k.a. 14-8H) reacts with both non-reduced and reduced LTP/CETP. Unlike clone 3-11D (3-9F; Cat. No. MABS1219), clone 14-8F (14-8H) did not block the lipid association of LTP/CETP with microemulsion, while both clones effectively blocked LTP/CETP-mediated triglyceride (TG) transfer from lLDL to HDL and from HDL to to apoB-containing lipoproteins (Ko, K.W., et al. (1994). J. Biol. Chem. 269(45):28206-28213).

Detect LTP/CETP using this Anti-LTP/CETP Antibody, clone 14–8F validated for use in Western Blotting, ELISA, Inhibits Activity/Function.
ELISA Analysis: Representative lots were conjugated with HRP and used as the detection antibody in combination with clone 3-11D (Cat. No. MABS1219) as the capture antibody for the detection of human plasma LTP/CETP by sandwich ELISA (Tosheska, K., et al. (2011). Bosn. J. Basic. Med. Sci. 11(3):169–173; Saito, K., et al. J. Lipid Res. (1999). 40(11):2013-2021; Sasai, K., et al. (1998). Clin. Chem. 44(7):1466-1473).
ELISA Analysis: A representative lot was conjugated with HRP and used as the detection antibody in combination with clone 3-11D (Cat. No. MABS1219) as the capture antibody for the detection of mouse plasma LTP/CETP by sandwich ELISA (Wu, C.A., et al. (2002). Arterioscler .Thromb. Vasc. Biol. 22(8):1347-1353).
ELISA Analysis: A representative lot, when conjugated with HRP and used as the detection antibody, showed pH-dependent LTP/CETP-binding activity with the maximum reactivity at a weakly acidic condition of pH 5 to 5.5 in sandwich ELISA applications (Saito, K., et al. J. Lipid Res. (1999). 40(11):2013-2021).
Western Blotting Analysis: A representative lot detected both rabbit and human LTP/CETP by Western blotting under either reducing or non-reducing condition (Ko, K.W., et al. (1994). J. Biol. Chem. 269(45):28206-28213).
Inhibits Activity/Function: A representative lot inhibited both rabbit and human LTP/CETP-mediated triglyceride (TG), but not cholesteryl ester (CE), transfer from low density lipoprotein (LDL) to high density lipoprotein (HDL) (Ko, K.W., et al. (1994). J. Biol. Chem. 269(45):28206-28213).
Inhibits Activity/Function: A representative lot effectively inhibited human LTP/CETP-mediated triglyceride (TG) transfer from high density lipoprotein (HDL) to to apoB-containing lipoproteins in human plasma. Clone 14-8F (14-8H) also inhibited cholesteryl ester (CE) transfer in the same assay by 40% (Ko, K.W., et al. (1994). J. Biol. Chem. 269(45):28206-28213).
