General description

Asialoglycoprotein receptor 2 (UniProt: P07307; also known as ASGP-R 2, ASGPR 2, C-type lectin domain family 4 member H2, Hepatic lectin H2, HL-2) is encoded by the ASGR2 (also known as CLEC4H2) gene (Gene ID: 433) in human. ASGPR is a type II membrane glycoprotein that contains a cytoplasmic domain (aa 1-58), a transmembrane domain (aa 59-79), and an extracellular domain (aa 80-311). Its C-type lectin domain is localized in amino acids 176-302. ASGPR is expressed exclusively in hepatic parenchymal cells where it serves in clearance of asialoglycoproteins from the plasma. It mediates the endocytosis of plasma glycoproteins to which the terminal sialic acid residue on their complex carbohydrate moieties has been removed. The receptor recognizes terminal galactose and N-acetylgalactosamine units. After ligand binding to the receptor, the resulting complex is internalized and transported to a sorting organelle, where receptor and ligand are disassociated, and the receptor is returned to the cell membrane surface. A soluble form of ASGPR (~ 35 kDa) has also been described and is termed as sH2a that is formed by cleavage in the endoplasmic reticulum of its precursor. Levels of ASGPR sH2a are significantly reduced under conditions of liver cirrhosis and fibrosis. Clone B9 specifically detects the secreted sH2a form of ASGPR in human plasma and in NIH3T3 cells transfected with human ASGPR sH2a. (Ref.: Shenkman, M., et al. (2013). J. Biol. Specificity

Clone B9 is a mouse monoclonal antibody that detects Asialoglycoprotein receptor 2 (ASGPR2). It targets an epitope within 12 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 12 amino acids from the C-terminal region of human Asialoglycoprotein receptor 2.

Application

Quality Control Testing

Evaluated by ELISA with recombinant human ASGPR protein.

ELISA Analysis (ELISA): Various dilutions of this antibody detected recombinant human ASGPR sH2a protein.

Tested Applications

Purified mouse monoclonal antibody IgG3 in buffer containing 0.1 M Tris-Glycine (pH 7.4), 150 mM NaCl with 0.05% sodium azide.
