Anti-iRhom1 Antibody, clone RHF1A 20A8

Inactive rhomboid protein 1 (UniProt: Q96CC6; also known as iRhom1, Epidermal growth factor receptor-related protein, Rhomboid 5 homolog 1, Rhomboid family member 1, p100hRho, RHBDF1) is encoded by the RHBDF1 (also known as C16orf8, DIST1, IRHOM1) gene (Gene ID: 64285) in human. iRhom1 is a multi-pass, homodimeric, membrane protein that is predominantly localized in the endoplasmic reticulum (ER). It is ubiquitously expressed, but higher expression is observed in brain. It regulates the maturation and activity of ADAM17, a sheddase of the epidermal growth factor (EGF) receptor ligands and TNF . It also controls the transport of ADAM17 from ER to the plasma membrane and removal of its inhibitory propeptide, and stability and degradation. It is reported to play a role in cell survival, proliferation, migration, and inflammation. However, it does not exhibit any protease activity on its own due to lack of amino acids that are required for proteolytic activity. iRhom1-deficient mice are reported to lack mature ADAM17 in the brain. Quality Control Testing

Evaluated by Western Blotting in 721.221 cell lysate.

Western Blotting Analysis: A 1:125 dilution of this antibody detected iRhom1 in 721.221 cell lysate.

Tested Applications

Western Blotting Analysis: A representative lot detected iRhom1 in Western Blotting applications (Weskamp, G., et al. (2020). J Biol Chem. 295(13):4350-4358).

Western Blotting Analysis: A 1:500 dilution from a representative lot detected iRhom1 in Mouse embryonic fibroblasts (MEF) lysates, but not in lysates with iRhom1 knockout (Courtesy of Katrin Moschke, Deutsches Zentrum für Neurodegenerative Erkrankungen (DZNE), Munich, Germany).

Clone RHF1A 20A8 is a rat monoclonal antibody that detects Inactive rhomboid protein 1 (iRhom1). It targets an epitope within 19 amino acids from the N-terminal region.
