General description
Myoferlin (UniProt Q9NZM1; also known as Fer-1-like protein 3) is encoded by the MYOF (also known as FER1L3, KIAA1207) gene (Gene ID 26509) in human. Myoferlin is a calcium/phospholipid-binding protein that is expressed in myoblast and endothelial cells and in cardiac and skeletal muscles. Low levels of myoferlin are seen in lung, and very low levels in kidney, placenta, and brain. Myoferlin plays a role in the plasmalemma repair mechanism of endothelial cells that permits rapid resealing of membranes disrupted by mechanical stress. Myoferlin contains C2 domains that participate in calcium-mediated membrane fusion events. It is also involved in endocytic recycling and has been implicated in VEGF signaling by regulating the levels of the receptor KDR. Myoferlin is overexpressed in breast cancer cells and its depletion blocks EGF-induced cell migration and epithelial to mesenchymal transition and reduces invasion through extracellular matrix (ECM). Immunogen
KLH-conjugated linear peptide corresponding to a sequence within the region between the second and the third C2 domain of human myoferlin.
Application
Immunocytochemistry Analysis: The undiluted hybridoma culture supernatant from a representative lot detected myoferlin in 4% paraformaldehyde-fixed, 0.3% Triton X-100-permeabilized C2C12 murine myoblasts.
Western Blotting Analysis: A representative lot detected myoferlin in C2C12 murine myoblast lysate and human skin fibroblast lysate. Antibody blocking with the immunogen peptide prior to Western blotting abolished the target band detection (Jaiswal, J.K., et al. (2007). Traffic. 8(1):77-88).

Biochem/physiol Actions
Antibody blocking with the immunogen peptide prior to Western blotting abolished the target band detection (Jaiswal, J.K., et al. (2007). Traffic. 8(1):77-88). Epitope is absent from human isoform 4 and mouse isoform 3, but present in all other human and mouse spliced isoforms reported by UniProt (Q9NZM1 and Q69ZN7).

Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.

Evaluated by Western Blotting in C2C12 cell lysate.
