General description

Chronic kidney disease (CKD) is caused by a variety of health factors but is often characterized by improper kidney function or abnormal kidney structure. Patients often start out asymptomatic but develop symptoms later in disease progression. Erythropoietin (Epo) is a hormone produced by the kidneys to induce red blood cell production in the body. Due to epo′s direct connection to the functionality of the kidneys, epo-deficiency anemia is a common problem that arises from CKD. Another complication that commonly affects CKD patients is renal fibrosis, a condition in which the kidneys become further damaged due to the excess production of extracellular matrix (ECM). This excess ECM production originates in myofibroblasts found in the peritubular interstitia. These dangerous myofibroblasts most likely originate through the transformation of tubular epithelial cells, capillary endothelial cells, migratory bone marrow cells, and interstitial fibroblasts. Interstitial fibroblasts are thought to be a major contributor due to the inflammation signaling response activated by nephron injury. However, as interstitial fibroblasts undergo myofibroblastic transformation, epo-production capabilities are inactivated. The general lack of appropriate cell models has made it difficult to understand epo-producing cells which also hinders the ability to understand underlying mechanisms of renal fibrosis. The SCC482 Replic Mouse Renal Cell line represents an important cell model in understanding the molecular pathophysiology of epo-producing cells. The cell line is derived from mouse renal epo-producing cells which were isolated and immortalized by exogenous expression of oncogenic H-RAS. The cells retain tdTomato expression which was utilized for isolation and cultivation. However, the Replic cells have lost their ability to produce erythropoietin due to continuous DNA methylation in the Epo gene promoter. The unique myofibroblastic features such as high TGFβ and the epo-producing defect of the cell line allows for a more thorough understanding of kidney fibrosis, renal anemia, and molecular CKD pathology.SourceDerived from kidneys of ISAM-REC mice which were later infected with lentivirus vector expressing G12V oncogenic mutant of human H-RAS. Application

  • Each vial contains > 1X106 viable cells.
  • Cells are tested negative for infectious diseases by a Mouse Essential CLEAR Panel by Charles River Animal Diagnostic Services.
  • Cells are verified to be of mouse origin and negative for interspecies contamination from human, rat, Chinese hamster, Golden Syrian hamster, and nonhuman primate (NHP) as assessed by a Contamination Clear panel by Charles River Animal Diagnostic Services
  • Cells are negative for mycoplasma contamination.

Features and Benefits

Replic renal cells have unqiue myofibroblastic features such as high TGFβ and aide in the understanding of kidney fibrosis, renal anemia, and molecular chronic kidney disease pathology.

Preparation Note

Replic cells should be stored in liquid nitrogen. The cells can be cultured for at least 10 passages without significantly affecting cell marker expression and function.
